  • Zymo DCC or Qiagen QIAquick for ChIP?

    Hi all,

    I'm just wondering if anyone has experience with the Zymo's and Qiagen's colums/kit in purifying ChIPed DNA.
    I saw that Zymo has a specific ChIP DNA purification kit and it said its binding buffer tolerates =< 1% SDS. The QIAquick PCR purification kit obviously does not say it's for ChIP nor the extent of SDS tolerance, but is mentioned/used more frequently in many protocols.
    I'm used to used the Qiagen kit but am thinking to switch if there's better ones out there. Is the Zymo kit a better choice?

    I am planning on a TF ChIP-Seq in plants which involves repeating several large-scale ChIPs and pooling the ChIPed DNAs at the end. So, I'm thinking to elute ChIPed DNAs from the columns with Water (instead of EB/TE) in each ChIPs and pool them by SpeedVac. Does it sound like a good plan?

    We've always used Zymo DCC columns for ChIP DNA with no problems. We just use the standard DCC-5 kits, though I imagine the specialty ChIP ones would be even better.
