    hi there!
i´ve read about the ESTexplorer, which is a web-based tool for pre/processing and annotating ESTs. so i tried it at http://estexplorer.els.mq.edu.au/estexplorer/ (i used the provided test-data) - but nothing happened! for 4 days now there is no progress in the status-report (still: "uploading data"). does anyone have experiences with this tool? does it really work?

    Thanks. Triticum.

My experience (about 18 months ago) was that when it worked, the progress was quite fast. However, I also had a similar experience to yours (no progress for days at one of the steps). I suggest you try resubmitting your sequences. Also, if you have another way to run CAP3 (or equivalent to build contigs), the annotation steps are more efficiently run directly with Blast2Go (http://www.blast2go.org/).

thanks a lot for your information. may be it would really be better if we use blast2go directly. i am just concerned about the computing-ressources, because we want to use 454-sequencing and so will have a huge amount of data. i thought that uploading to ESTexplore would provide a better performance for computing this amount of data, because currently we do not have a dedicated server for these calculations.

        Current Approaches to Protein Sequencing
        by seqadmin

Proteins are often described as the workhorses of the cell, and identifying their sequences is key to understanding their role in biological processes and disease. Currently, the most common technique used to determine protein sequences is mass spectrometry. While still a valuable tool, mass spectrometry faces several limitations and requires a highly experienced scientist familiar with the equipment to operate it. Additionally, other proteomic methods, like affinity assays, are constrained...

        Strategies for Sequencing Challenging Samples
        by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
