Is it possible to make a good whole blood RNASeq library without depleting globin?
I am just starting my adventure with RNASeq. I want to make an adult zebrafish whole blood RNASeq library, but I need total expression data and can not (would strongly prefer not to) exclude globin. Will it be challenging to get a good read from non-globin-depleted sample? Is it just a matter of increased number of reads? I found a paper dealing with embryonic zebrafish whole blood library where they didn't mention any globin depletion, but I'm not sure how it relates to an adult sample. Can I expect them to be similar in globin content? I have heard that globin content may differ between species, so I specified my model.
