  • SOLiD library on Ion Torrent

    Hello,
    does anyone of you already has experience with putting a SOLiD (small RNA) library on the Ion Torrent?
    Which kit or protocol did you use or do you recommend?

    Hi GenomicIBK,

    You can't actually run a SOLiD library on the PGM since the adaptor sequences on the beads are different for each platform. You'll have to make a new Ion library from your RNA.

    You can use the Ambion Ion Total RNA-Seq Kit to make your Ion small RNA library. (The protocol is very similar to Ambion's SOLiD Total RNA-Seq kit.) This kit will allow you to make a library that is compatible with the Ion PGM bead prep.

      It seems I was a bit unclear with my explanation, sorry. My question was, if I already have a SOLiD library, how can I sequence it on Ion Torrent (e.g. as quality control for a multiplexed SOLiD library).
      A colleague gave me the tip to simply ligate the primers for Ion Torrent onto my SOLiD library, what I will do now.
      I´ll let you know when I have a result.

        Seems like if you were planning to do this a lot you would want to create Ion Torrent - SOLiD "conversion primers". That way you could make your normal SOLiD library, amplify it with your conversion primers and you would have a Ion Torrent library.
