Hi folks. Could someone tell me that how indexes and sequencing primers are added in TruSeq Custom Amplicon v1.5 library preparation? By ligation (to the end of custom probe 1/2) or by base pairing (hybridize to custom probe 1/2)? Any answer appreciated!

Judging by the picture you joined to your message, they are added during the PCR step.
Indexing during PCR is rather common ine Illumina's protocols especially when for amplicons and when using Nextera. Last edited by huguesparri; 01-18-2017 at 07:41 AM. Reason: typo |
