  • Can I use same adapters for PE and single read?

    Greetings everyone,

    I am a novice and my question would probably seem trivial to most of you

    I haven't decided yet whether I need single read or paired-end read sequencing. I would like to start from a single read (cheaper ).

    Therefore I wonder whether I can use adapters for Paired End reads for making a library that would be sequenced by single read protocol?

    Yes use Illumina's TruSeq adapters. You can get the sequences from Illumina tech support.

    Or as a beginner you are probably better off buying a kit. The price per library for most kits isn't more then buying the reagents separately. But it depends on your application. Without more specifics there isn't more to be said.

      Paired End DNA oligonucleotide sequences
      PE Adapters
      5' P-GATCGGAAGAGCGGTTCAGCAGGAATGCCGAG
      5' ACACTCTTTCCCTACACGACGCTCTTCCGATCT
      PE PCR Primer 1.0
      5' AATGATACGGCGACCACCGAGATCTACACTCTTTCCCTACACGACGCTCTTCCGATCT
      PE PCR Primer 2.0
      5' CAAGCAGAAGACGGCATACGAGATCGGTCTCGGCATTCCTGCTGAACCGCTCTTCCGATCT
      PE Read 1 Sequencing Primer
      5' ACACTCTTTCCCTACACGACGCTCTTCCGATCT
      PE Read 2 Sequencing Primer
      5' CGGTCTCGGCATTCCTGCTGAACCGCTCTTCCGATCT

      I plan to request reads from ACACTCTTTCCCTACACGACGCTCTTCCGATCT
      would that be OK?

        Yes you can use them for single read sequencing.

        The single read sequencing primer is the same as the paired end read 1 sequencing primer.
