  • Combining MiSeq cartridges to get longer rads

    Hi all,
    I would like to combine three 50 reads cartridges (we have half of freezer of expired ones) to get 2x75bp reads. I know it is possible and that it will work and used to have publication where they were making 1200bp reads on MiSeq. But I cannot find it now, so I would like to ask what wells I have to combine and if I have to disassemble the cartridge (not easy) or if I can just puncture the tinfoil and combine them using pasteur pipette.

    First get an RFID override code...

    1. you would need an RFID override code from the Illumina support.
    2. The pasteur pipette should work ok (make sure it is sterile and protease/DNA'se free!)
    3. If trying longer reads (>250bp), try lowering the clustering density to get a better quality and lower dephasing...
    4. Keep the room with the miseq on a cooler side - 18 degrees or so.
