  • Are there alternatives to the Sanger sequencing for Big Dye Terminator kit?

    Hi, All

    As far as I know, there are two alternatives from Nimagen(Brilliantdye) and McLab(Brightdye). Does anyone have experience/feedback on these two or other alternatives?

      I realize this is an old thread, but does anyone use Promega's ProDye and can the volume used be cut down to 1/8th or 1/16th, as we used to do with BigDye 3.1?

        I think it should be possible to dilute most big-dye chemistries down to 16X - 32X fold, provided you add some DNA polymerase (Like thermo DreamTaq),
        use 10ul of Taq polymerase for 400ul of digdye/buffer mix for 96 reactions.

        Also it can help resque big Dye mix, that had been held in customs for a few weeks at room temperature when being shipped to some third world country...

        Also make sure the quality of your DNA is good or use PCR products for sequencing.
