  • TruSeq Custom DNA kit

    Hi all,

    I am looking for advice from anyone who has experience of using the TruSeq Custom Amplicon Library Preparation kit please! Part of the protocol requires a 96well heat block that will cool samples from 95C to 40C over 80 minutes. We don't have the reccomended incubator, and Illumina Tech Support have said we can't use a thermal cycler. Our hot blocks take 240 minutes to cool to 40C - way to long for the samples to work. Has anybody used this kit with anything other that the recomended Scigene Hybex microsample incubator?? - don't want to have to spend out to test the kit out.

    I don't have a Hybex either. I've programmed a thermal cycler to cool to 40 over 90 minutes or so (drop 0.1 degrees every 10 seconds) for 550 cycles. Seems to work fine for my panel (~550 amplicons). Also, Illumina tech support said the ACD1 process control results were fine.

      moggz101- Did you end up getting the Hybex? I'm trying to avoid some of these hidden costs associated with Illumina library prep kits but it looks like this incubator is used in a few of the capture based kits.
