  • NovaSeq metatranscriptomics

    Hi all,

I was wondering if anyone has experience with (a) metatranscriptomics run(s) on the NovaSeq 6000?
Our preferred company switched from HiSeq to NovaSeq. We are planning to sequence about 110 soil samples.
Some years ago, we used the BiooScientific NEXTflex™ Rapid Directional mRNA-Seq Kit for a HiSeq 2500 run.
I guess NEXTFLEX® Rapid Directional RNA-Seq Kit 2.0 or the NEXTFLEX® Rapid Directional qRNA-Seq™ Library Prep Kit are the current incarnations. Are these compatible with NovaSeq? Are there alternatives you could suggest?

    Thanks!

    Best regards

    Does anybody have any suggestions?

In addition, is it possible to multiplex >100 samples on a NovaSeq using existing tags?

    thanks

Yes it is possible to do 384 (and many more if you do custom designs).

The IDT® for Illumina®-Nextera™ DNA UD Indexes Sets A, B, C, and D Indexes offer up to 384 unique dual indexing that enables accurate assignment of reads and efficient use of the flow cell. These unique dual index codes use 10 base pair codes.

        Current Approaches to Protein Sequencing
        by seqadmin

Proteins are often described as the workhorses of the cell, and identifying their sequences is key to understanding their role in biological processes and disease. Currently, the most common technique used to determine protein sequences is mass spectrometry. While still a valuable tool, mass spectrometry faces several limitations and requires a highly experienced scientist familiar with the equipment to operate it. Additionally, other proteomic methods, like affinity assays, are constrained...

        Strategies for Sequencing Challenging Samples
        by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
