  • Strange peak after cDNA enrichment

    Hello,

    Now I am trying to do RNA-Seq for DEG analysis between different tissues of plant root.
    So far, I succeeded to extract total RNA (1-5 ng/ul in 10 ul, RIN ~ 7.0) from cryosectioned samples by using LCM technique.
    I tried SMARTer kit and population Quartz-seq for cDNA synthesis and amplification, but both methods produced a strange peak which is longer than 10kb (High sensitivity DNA kit result attached). I have no idea what this peak is.

    Has anyone seen this kind of huge peak? or Do you have any good ideas to make library using this sample?

    Hi pjtkkw!

    I am preparing single cell cDNA library using Quartz-seq method. Now I have a problem, Aglient 2100 data showed that there was big peak at ~600bp. I used 21x cycles in the PCR. I find that your Aglient 2100 data had the same problem. Did you fix this problem?
