  • Number of samples on a 316 chip?

    I would like to sequence the whole genome of several E.coli mutants, genome size 5.3MB, to look for single snps. (Using Ion torrent PGM) I have 8 samples in total, would it be realistic to run them all on a 316 chip?

    Any advice would be great, I'm completely new to this!

    The PGM 316 chip can generate around 1Gb of data. So for 8 samples with 5.3Mb genomes, you’ll get roughly 20X coverage of each genome. It depends on what you’re trying to do with the data, but that coverage sounds a little low. You could either bump up to the 318 chip to double your coverage, or run only 4 samples per 316 chip.
