  • Evaporation During End Repair on Automated Platform

My company is trying to run A-tailing and End repair on an automated liquid handler. During one of the steps, the temperatures get up to 65C and causes evaporation of the sample.

When done manually, a plastic seal is placed over the plate like in PCR. Since we do not have a plate sealer on deck, we have been looking into various types of lids to prevent the samples from leaving the well.

We are trying to avoid using oil to prevent the evaporation because that introduces more variables that we have to program into the liquid handler. If we can find a way that works with different types of lids or something else that works just as well, the setup would work perfectly.

Anyone experience this problem and found a way around it?

Yes we did: we are sealing the plates manually. So far, it's the easiest and surest way we found to prevent evaporation.
Sorry if it's not the answer you were hoping for :s

      Strategies for Sequencing Challenging Samples
      by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
