  • Data analysis Exome sequencing

    Hello everyone, i have performed exome sequencing, but when i extract the raw data from the software of ion torrent, it came out to be only 1.7Mb, ISPs loading was only 28% and only approx 2 million reads were obtained. can anyone tell me what is the problem with the data.??
    one flaw i can share with u, during the hybridization of the amplified library the volume of solution decreases, as it must be supposed to be nearly 13ul (initial 15ul) but after hybridization of 71 hrs it came out to be less than 7ul. Can this be the reason for less loading of ISPs?

    While someone here may have an explanation it may be best to talk with Ion tech support to hear what they have to say about a specific observation like this.

      Hello!
      The title of your post mentions exome sequencing, and you refer to the Ion Proton, but I'm not aware of any 71-hour hybridization step on the official protocol.
      Could you point me to the protocol you're using? Thank you!
