  • running RTA offline

    Hi,
After some data of HiSeq2500 run were accidentally deleted, I have the following files for each tile in the 'Processed' folder:
    *.errorMap
    *.qms
    *.stats
    *.FWHMMap
    *.align
    *.bqm
    *.readErrors
    *.bqm
    *.clocs
    *.control
    *.ctr
    *.filter
    *.tempdensity
But I don't have *.bcl or *.cif files.
Is there any option to create bcl files for this run? My RTA version is 1.18.66.4. I saw that there is an option to run RTA offline (OLB, off-line base-calling), but when I run it (double-click the RTAstart.bat file) I get the message that bcl files are already generated.
    Thanks for helping.

No, without .bcl or .cif files you're stuck.

      Strategies for Sequencing Challenging Samples
      by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
