  • 96-Well Manual Hyb Wash using Agilent SureSelect XT

    I am using Agilent SureSelect XT for targeted enrichment. The provided protocol describes preforming the hyb wash in 1.5ml tubes. I will be needing to prepare 48 or more samples at a time. I need to do this manually as we do not have automation in our lab yet. Has anyone preformed the hyb wash in a 96-well plate? I assuming you would need decrease the volume of buffer and increase the number of washes, but any help or example where this has been successful would be greatly appreciated!

    Yes-I have done it.

    Use a wash volume of 150ul and wash it 5x instead of the 3x they recommend.

    Just be careful as 150ul can come close to the top and you don't want cross contamination.
