  • CBot vs. Cluster station

    Hello:
Illumina says the cBot will give slightly higher cluster numbers than the cluster station with the same library concentration. Has anyone seen this? How much higher? We are using 10 pM for the library currently, to get 350,000/mm2 on the old cluster station.

    Thanks for your help!

I guess it depends on your library quantification, too. We constantly get 400K/mm2 with 6.5 pM input (on cBot).

The cBot is definitely more consistent and gives higher cluster numbers. We get more like 500 to 600K/mm2 PF clusters

I get about that for passing filter clusters with RNA-seq libraries on the cBOT.

          Strategies for Sequencing Challenging Samples
          by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
