  • Homemade SPRI beads- hydrophobic or hydrophilic beads?

    We want to make our own Ampure beads for DNA cleanup and size selection.
    We think we need the Sera-Mag Carboxylate-Modified SpeedBeads, but they come in two types; hydrophilic and hydrophobic. We are thinking the hydrophilic is what we need, but here they link to the hydrophobic variety.

    Hi,

    I'm using Fisher #09-981-123 (GE Life Science 65152105050250) according to the protocol posted by Ethan-omics: https://ethanomics.wordpress.com/201...pure-xp-beads/

    No problems so far, the selfmade beads are working perfectly.

      Those are the same ones in the protocol JBKri is looking at, since we just copied the purchasing information when we improved on the other aspects of Faircloth & Glenn's protocol.

      JBKri, why do you think they should be hydrophilic beads? Or, why do you think the carboxyl coating is hydrophobic?

        Thank you both! A colleague uses the homemade beads, and at one point there was some confusion about the beads; we were mistakenly given a product code for hydrophilic beads.
