  • Questions regarding human saliva gDNA extractions for NGS applications

    Hi! I have some questions regarding human saliva gDNA extractions that I would hope fellow scientists that are experienced in the area could assist me. To aid you with some of the questions, here are the requirements:
    • My institution has Illumina NextSeq 500 machine for Exome Sequencing
    • The project is about pharmacogenomics
    • The sample size is 150 people
    • The sample is in the form of human saliva
    • The downstream application that will be used on the extracted gDNA is exome sequencing.


    The questions are:
    • What kind of DNA extraction kits do you recommend me using for this sort of project? I need the kits to be affordable since the budget for the project is very tight.


    Questions for people who have some experience using Qiagen's modified DNeasy and/or Blood Mini Kits for human saliva samples:
    • What are the expected yield for the modified DNeasy kit for saliva samples?
    • What are the precautions and steps that we should pay attention to when using these modified kits?


    Apologies if this is the wrong place to ask this question. I'm quite new and I rarely visit this website. Any additional advice is appreciated.
