  • MALBAC single cell WGA, any experience?

    We have a difficult-to-grow sample of which we'll be doing genomic sequencing. We may need to do whole genome amplification of single cells. We've used Qiagen REPLI-g, but MDA has its disadvantages. Has anyone tried MALBAC? Did you follow the protocol in the original paper, or modify it, and how did it go for you? Or has anyone tried this MALBAC WGA kit from Yikon Genomics

    This is the first I've heard of MALBAC. However, there are some MALBAC reads (Human Sperm Genome Sequencing at Harvard) in the SRA that may yield insights into the nature of MALBAC.

      if the sample is human, then the best WGA for mutation detection and Copy number analysis is Ampli1 (menarini silicon biosystems)

        Malbac

        Though it may have improved by now, we had a horrible time with it. Kit did not arrive for weeks, was missing half the reagents when we did get it.
        Tech support was non-existent and then the sequencing data was not good.
