  • Illumina insertions deletions

    Hello,

Is it safe to assume that a higher coverage tends to resolve the nt-substitutions that can occur in Illumina sequencing?

  • #2
Yes, as long as the higher coverage comes from higher input of source material the errors should be mainly random, and suppressed by more data. If you raise the coverage by amplifying your sample with more PCR cycles, then you risk to introduce more substitutions the higher "coverage" you have.
