  • HTP RNA extraction from plants?

    I need to extract RNA from ~1200 leaf and stem tissues. So far I've found a protocol using the qiagen 96well RNA easy kit for plants. Is that the only high throughput RNA kit that has been used for plants or are there others that might work better? I have an epmotion liquid handler but am comfortable programing it myself if there isn't a program for a particular kit.

    Do you mind handling phenol in a 96-well format? If not, you could also try using Zymo's Direct-Zol 96 RNA kit -- I've been meaning to test it for tissue/cell RNA extractions for a while. If you have access to a TissueLyser or the equivalent for high-throughput lysis it could work quite well.

    Just FYI, Qiagen's RNeasy-96 plates give great yields but require access to a centrifuge with buckets of a very specific format. I've had to tweak the protocol somewhat to get rid of all wash buffer before eluting my RNA.

      I'd prefer not to use phenol - there's no way to set up the robot in a hood for one.
