  • ampure vs alternatives

    I'm about to start running 100s of libs through double spri cleaning. In the past I've used ampure and made my own PEG solution which allowed me to use ~1/5 the recommended amount of Ampure. (single clean up for ~$0.25). This would mean $0.50 double clean ups which isn't unbearable but since I'm starting at the beginning for optimizing this protocol (and we don't use Ampure for any other lib preps) I feel like this is the time to look around for cheaper ways to do it. I'm hesitant to go fully homebrew because we're a core facility and I worry about clients balking at us not using a kit.

    So please anyone who does large numbers of preps-how are you controlling bead costs?
    Microbial ecologist, running a sequencing core. I have lots of strong opinions on how to survey communities, pretty sure some are even correct.

      I've had great luck using these:
      Sera-mag SpeedBeads (Fisher#09-981-123)
      You basically just wash them, add your own PEG solution, and bingo! you've made AMPure beads for almost zero dollars.
