  • TruSeq PCR Prep Box - DNA/RNA

    Hi all,

I wonder is there any difference between TruSeq DNA PCR Prep Box and TruSeq DNA PCR Prep Box?
Looks like we accidentally used RNA box instead of DNA for enrichment of DNA libraries. It worked well but now I'll have to use DNA box for RNA libraries because I don't have a new RNA prep kit at this moment. Will this work? Probably, some modifications should be done? (I know that DNA adapters have higher concentration than RNA ones, so I guess that primers from DNA PCR box can have higher concentration too.)

    Thanks!
    Maria.

    Hi!

I guess it should work, as we have received Truseq RNA kits with the Truseq DNA PCR box... so, either Illumina had a problem with the shipment and we received something we were not supposed to get, or both truseq RNA and DNA PCR boxes are the same (although I would expect a difference in primer concentration, as you said).

Yes, we also received Truseq RNA kits with the DNA PCR box....

Thanks for sharing your ideas and experience. I ended up with doing no modifications and this worked pretty well. I was afraid that I could have a lot of adapter dimers but this does not seem to be the case.

Recent Advances in Sequencing Analysis Tools
          by seqadmin

The sequencing world is rapidly changing due to declining costs, enhanced accuracies, and the advent of newer, cutting-edge instruments. Equally important to these developments are improvements in sequencing analysis, a process that converts vast amounts of raw data into a comprehensible and meaningful form. This complex task requires expertise and the right analysis tools. In this article, we highlight the progress and innovation in sequencing analysis by reviewing several of the...

Essential Discoveries and Tools in Epitranscriptomics
          by seqadmin

The field of epigenetics has traditionally concentrated more on DNA and how changes like methylation and phosphorylation of histones impact gene expression and regulation. However, our increased understanding of RNA modifications and their importance in cellular processes has led to a rise in epitranscriptomics research. “Epitranscriptomics brings together the concepts of epigenetics and gene expression,” explained Adrien Leger, PhD, Principal Research Scientist...
