  • PromethION and Barcoding

    Dear all,

    we will get a PromethION machine surprisingly soon, and you know, rumours are fast, so all of our researchers are already standing in our lab to have something sequenced.

    The question is now, how to barcode all the DNA since there is no barcoding kit available from ONC?

    I guess someone is already doing it, no?

    Are there maybe already protocols available?

    I have no idea what you are going to sequence, but as you said barcoding kits are not available at this moment. However, I've been told that these would get released "in the following weeks". You could definitely sequence with barcodes, you also have to make sure your basecaller supports it.

    I don't know about the size of the genomes you are sequencing, but perhaps take it easy for the first few runs until you get more comfortable with the system.

      There is a discussion thread in Nanopore community. It can be accessed by logging in and searching for "PromethION barcode".


      It has been tried successfully but Nanopre is working on an optimized protocol and currently dose not support this application.
