Development of Index : project of a desperate student

I am a student and I have to do a project on multiplexing, but I can not understand some things.

I would like to ask you for help, I have a question.

my teacher wants that I explain how the indexes are built for multiplexing, but I can find no information on the subject.

can you please explain me how to build indexes (6nt) when we have multiple samples to be analyzed to reorder them and no information is lost (or contamination)

how to vary the sequence of indexes (indexes for 6nt) for several samples?
