Hi, I am about to start a resequencing project on the HiSeq 1000 searching for rearrangements in a bacterial genome.
This is the 1st time I am going to use the mate pair approach, so I am wondering if any of you can share their experience about the protocol especially concerning crucial steps, tips, tricks, and any particular problem you may have had during the library prep. I had a bad experience with paired end library prep on 454 (8-20kbp) last year so I am concerned about starting this new protocol. Thanks to the folks who will share their experience ![]() |
