Does anyone have any experience with library preparation of chromosome sorted DNA? The main issue is that we are starting with only 120ng of material. The good news is that we don't have to do any hybridization/capture, so we just have to repair the ends, add the A, ligate the adaptor and PCR the library. Any guidance from people who have sequenced this type of material would be very much appreciated.
