Prepping a sample for a GS Jr run. Realized the Enrich and Seq primers got left out overnight at room temperature. We've got calls (and emails) in to Roche tech support, but it's been more than an hour, and now we're just sitting here twiddling our thumbs. Anyone know if these reagents are ruined? Or are they like real primers and likely fine?

I don't have any direct experience with your issue, but I suspect they will be just fine. At this point, you have nothing to lose by going ahead with the enrichment. If the enrichment primer has gone bad, you will get poor results from your enrichment and it will be clear after the washes. If the enrichment primer is okay then the sequencing primer should be fine as well. I'd say go ahead try it, then come back here and let us know how it worked out. -

Data variability is still an issue in sequencing technologies despite the advances in reproducibility and accuracy of these platforms. But the problem does not originate in the sequencing itself, but in the previous steps, before the sample reaches the sequencer.
The first step is collection, followed by preservation and sample preparation for analysis. Most scientists overlook those steps, but not being careful might just be skewing the experiment’s results.
