    Fragment size of genome re-sequencing

    Hi All,

    I am going to do a genome re-sequencing, in order to find the mutant gene in Arabidopsis. Does anybody know what size of DNA fragment should be fine for this experiment? Any advice would be grateful!

    What sort of mutation you are looking for? Is it a point mutation? Are you expecting only one or multiple mutations?

      Yes, we are looking for a point mutation. From genetic study, it is probably a single mutation.

        In this case fragment size will not have any effect on mutant detection, only coverage will be important. Any standard Illumina DNA library should be OK. The paper in this link describes a similar approach and may be of interest to you:http://www.nature.com/nbt/journal/v3.../nbt.2095.html

        You might need to follow papers citing this one to see if simpler method has been published since this.
