    NebNext Adaptors for illumina

    Hi all,

    anyone know wchich adapters are better for whole genome sequencing, I'm thinking about NebNext loop adapters. Are they better than standard Y shape adapters.?

    I cannot think of any advantage. They add one extra step for protocol and most likely NEB uses them to avoid IP issues.

      I read in some article that loop adapters are more effective during ligation and we can expect more mapped reads to reference genome.

        I could imagine that they are somewhat more efficient (as NEB claims) thanks to the adapters being a single molecule. Perhaps they have advantages for low input applications because of this.
        They have the drawback, though, that you can't use them for PCR-free libraries. If you have enough DNA, PCR-free libraries would my suggestion.
