    MiSeq Micro/Nano Kits v2 for 16S?

    Hi guys,

I was wondering if anyone have used MiSeq Micro v2 (2x150) or Nano Kits (2x150 or 2x250) for 16S metagenomics? So far, I have experience only with standard v2 kit 2x250 and it was working well. However, now I'll have only 20 samples to sequence and I was wondering if it is worth trying smaller flow cells to reduce the cost? Also, I wanted to ask what coverage are you using then?

    Cheers

If I recollect the cost advantage was not that great with nano (perhaps micro) kits since both use normal flowcells (with some blacked-out regions, if I remember right). Have not looked recently but that may still be the case. Have you looked at the yields/unit cost?

They should work but I've never tried them either. 2/3 cost for 1/10 sequences means that I'll just sit on samples till I get enough in to run the regular size kit.
Microbial ecologist, running a sequencing core. I have lots of strong opinions on how to survey communities, pretty sure some are even correct.
