Protein Homology

Hello

I am working on a recombinant protein, and I want to find out its homologs. Using the dali server first and picking up the proteins having maximum simalarities and then doing the similarity search using cluster omega is the right way?or i can randomly pick the members of protein family to which my protein belongs and then use cluster omega?

The Dali server searches for homology in solved 3D structures.
I think a BLAST search should be good enough for your needs.
