    Improving PCR-free NGS library preparation

    PCR-free NGS libraries reduce bias, improve coverage of difficult areas, and enhance the detection of sequence variants. However, the current PCR-free NGS library prep kits tend to have low sequencing yields than that of PCR amplified libraries because:

    • Full libraries are mixed with partially ligated libraries and un-ligated DNA fragments, thus making it difficult to optimize library quantity due to inaccuracies in library size and amount.
    • Partially ligated libraries and un-ligated DNA fragments reduce cluster efficiency.

    In order to address this issue, BioDynami has developed the PCR-free NGS DNA Library Prep Kit.



    Our PCR-free kit effectively removes partially ligated libraries and un-ligated DNA fragments, resulting in only fully ligated DNA fragments in the final library. The maximal sequencing yield can be reached since cluster density is optimized based on accurate library sizes and amounts. Also, there is no clustering interference caused by partially ligated libraries and un-ligated DNA fragments.

    Data variability is still an issue in sequencing technologies despite the advances in reproducibility and accuracy of these platforms. But the problem does not originate in the sequencing itself, but in the previous steps, before the sample reaches the sequencer.


    The first step is collection, followed by preservation and sample preparation for analysis. Most scientists overlook those steps, but not being careful might just be skewing the experiment’s results.
