    Crispr library which gene?

    Hi,
    we're planning to do several Crispr runs to answer several question relating to translational machinery.

    I was wondering whether there are prepared libraried for such experiments, or if one need to create his/her own library.
    If I would like to answer different questions, does it make more sense to use several different libraries, or better to use one bigger, with more gene/sgRNA library.

    The bigger question is how do I choose which genes should go into the library. Are there any parameters we should consider, or just take a genome-wide library and hope the gene we want are in it?

    Data variability is still an issue in sequencing technologies despite the advances in reproducibility and accuracy of these platforms. But the problem does not originate in the sequencing itself, but in the previous steps, before the sample reaches the sequencer.


    The first step is collection, followed by preservation and sample preparation for analysis. Most scientists overlook those steps, but not being careful might just be skewing the experiment’s results.
