I'm working on a proposal to study non-coding RNA expression, mRNA expression and splicing in clinical samples of autoimmune diseases. I know that we can get some information about microRNA from RiboMinus-prepared RNA-Seq data, but the best way to study microRNA is through separate small-RNA sequencing.
If we do both small-RNA sequencing and RNA-Seq, can we use the same amount of total RNA for sample preparation? Probably we can select small RNA and long RNA based on their size difference. If we have to do separate sample prep, how much more total RNA do we need for small-RNA sequencing?
