How can estimate the required coverage

Hi all:

I do not know how to answer a very simple question. I request you to help me.

How do I know what is the required coverage for a 2 KB region. Someone asked me what is coverage needed to find a very low frequent mutation (10-5 10 to power -5). He wants to find mutation in one locus in a 2KB region. He asked me what is the coverage he should aim for?

To be honest, I do not know how to answer that question. Could someone help me here please.
