    DNA Extraction from Bone

    Has anyone tried to extract DNA from bone and then successfully create an Illumina Library from it?

    We have such material from a customer and it seems the concentrations are correct, and it shears appropriately, but we have tried twice (unsuccessfully) to create a library.

    The customer says she used the "kit from Covaris".

    Any thoughts?

    I did. There are whole bunch of methods described in detail in papers from Paabo's lab. The most important is to use fluorescence-based DNA quantification methods, which preferentially detect dsDNA. I cannot comment more as DNA quality depends on too many factors when dealing with bones. The sample may have too much inhibitors, degraded, etc.
